Sofija Hohencolerna bija Prūsijas hercogistes princese.
Prūsijas hercoga Albrehta Frīdriha trešajai meitai vārds esot dots par godu viņas vecvecmāmiņai Sofijai Jagellonei (1464–1512), kuras tēvs bija Polijas karalis Kazimirs IV Jagello, bet māte – Elizabete fon Habsburga. Tādējādi hercogienes senču vidū bija ne tikai Prūsijas, bet arī Polijas-Lietuvas un Austrijas valdnieki.
Kāzas notika Kēnigsbergā, sievu Vilhelms pārveda uz Kuldīgu tikai pēc vairākiem mēnešiem.
1609. gada 22. oktobrī apprecējās ar Kurzemes hercogu Vilhelmu.
Sofija mira nepilnas četras nedēļas pēc prinča Jēkaba dzimšanas, proti, 1610. g. 24. novembrī (v.s.)
